B vitamins are a group of eight essential vitamins with several critical functions in the body. Vitamin C is perhaps historically best-known for the treatment and prevention of scurvy, though it has many other roles in the body. For example, vitamin C catalyzes or is a cofactor in eight enzymatic reactions involved in the synthesis of collagen, the nutrient carnitine, and several neurotransmitters necessary for the proper function of the brain.<\/p>
In the contemporary context, vitamin C is best known as a potent antioxidant. In addition to its intrinsic activity as an antioxidant, vitamin C has been shown in in vitro trials to regenerate alpha-tocopherol (vitamin E). As an antioxidant, vitamin C combats the detrimental effects of a group of compounds called radical oxygen species that, when produced, degrade the lipid membrane of the cell and may cause internal damage. By \"scavenging\u201d these free radicals, vitamin C and other antioxidants form a defense against excess cellular damage.<\/p>
Vitamin D, in its various forms, is not an essential nutrient \u2013 as it is produced endogenously within the body, as a reaction to the skin\u2019s exposure to the sun \u2013 and thus not commonly considered a vitamin. Despite its technical classification, vitamin D (a hormone), is nevertheless an import and biologically-active compound, necessary for the calcium homeostasis and metabolism, along with increasing the absorption of magnesium and phosphate.<\/p>
Vitamin E is another technical misnomer, as the term refers to a group of compounds known as tocopherols with eight biologically-active constituents: alpha-, beta-, gamma-, and delta-tocopherol and alpha-, beta-, gamma-, and delta-tocotrienol. Alpha-tocopherol is the principal bioactive in humans, though the presence of other tocopherols has been linked to beneficial effects.<\/p>
As a result of its robust antioxidant activity, vitamin E assists in the maintenance and support of several physiological processes, though most notably sight and the immune system. Emerging research also suggests that the alpha-tocopherol form of vitamin E inhibits an enzyme known as protein kinase C, and therefore contributes to the healthy regulation of smooth muscle cell growth and differentiation.<\/p>
Critically, we have used the natural version of vitamin E (technically d-alpha tocopherol) and not the synthetic (dl-alpha tocopherol). The difference is a meaningful one: the single isomer of d-alpha tocopherol is easier and more efficiently absorbed and used in the body as compared to the eight isomers in the dl analog.<\/p>
While best-known as the main bioactive in milk, and a compound that contributes to strong bones, calcium has numerous physiological effects in the body \u2013 including mediating vascular contraction and vasodilatation, muscle function, nerve transmission, intracellular signaling, and hormonal secretion. Homeostatic regulation of calcium, which the body cannot produce, requires sufficient levels of vitamin D.<\/p>
When present in sufficient amounts, calcium is necessary for the adequate formation, function, and remodeling of both bone and teeth. Insufficient levels of calcium are associated with degenerative bone and joint disorders such as osteoporosis, wherein bone accretion slows or stops; thereby reducing bone mass and density and decreasing bone strength.<\/p>
Iodine is a trace element naturally present in certain foods, including kelp. Iodine is a critical component for fetal development, especially during early pregnancy, as maternal T4 (thyroxine) is the sole source of fetal thyroid hormone. After birth, especially if breast-feeding, iodine remains an important constituent of a balanced-diet to maintain adequate T3 (triiodothyronine) and T4 (thyroxine) levels in the mother and for the proper cognitive development in the child.<\/p>
As implied, however, the most important of iodine\u2019s functions is assisting the body to produce and maintain healthy levels of both thyroid hormones and TSH (thyroid stimulating hormone, released by the pituitary). The thyroid hormones, in turn, regulate several critical biochemical reactions, including protein synthesis, and, along with norepinephrine\/epinephrine, largely determine the body\u2019s metabolic rate.<\/p>
It is difficult to overstate the biological necessity of magnesium. Magnesium is a co-factor in over 300 enzymatic reactions that regulate essential physiological functions such as protein synthesis, glucose homeostasis, muscle and nerve function, and the maintenance and support of healthy blood pressure levels.<\/p>
Magnesium is also required for the use of the body\u2019s energy currency, ATP. To become biologically active, ATP must be bound to magnesium to form Mg-ATP. In addition, magnesium is necessary for the proper function of cells with calcium and potassium gated ion channels such as heart and muscle cells.<\/p>
In men, magnesium (along with zinc) is necessary for the maintenance of healthy levels of testosterone.<\/p>

Most notably, zinc is vital for immune function, protein synthesis, cellular division, and DNA synthesis. For immune function, zinc is required for the function of a group of immune cells known as neutrophils and macrophages, which eliminate pathological cells and their products from the body.<\/p>
With magnesium, sufficient levels of zinc are also necessary for the production and maintenance of healthy levels of testosterone.<\/p>
Copper is a trace element necessary for the production, function, and maintenance of a diverse range of compounds involved in physiological function.<\/p>
The brain, the heart, bone, and connective tissue specifically require copper or the enzymes it catalyzes to properly grow and differentiate their various cell types. Like many of the ingredients in Core MULTI, copper is also required for the homeostasis of other minerals, specifically zinc.<\/p>
Manganese is a required element for not only humans, but all living organisms. Manganese primarily acts as an antioxidant, specifically scavenging a radical oxygen species known as superoxide.<\/p>
Chromium is an essential, though little understood, trace element found in certain foods and industrial compounds. Only one of chromium\u2019s two forms, the form used in Core MULTI, chromium 3+, is biologically active.<\/p>
While not as well-defined as other trace elements, chromium\u2019s physiological actions nevertheless seem pivotal to the production and transmission of healthy levels of insulin. Emerging research also seems to suggest that chromium is directly involved in the metabolism of carbohydrates.<\/p>
Molybdenum is an essential trace element, involved in at least four enzymatic reactions necessary for urea excretion and a number of other physiological functions.<\/p>
There is extensive literature demonstrating that berberine is a potent AMPk agonist in skeletal muscle. Based on the pharmacological similarities between berberine and the anti-diabetic medicine metformin (with respect to AMPk activation), it is possible that berberine may promote an anti-catabolic state through increased expression of AMPk.<\/p>
The human data \u2013 in both healthy and disease state populations \u2013 demonstrating berberine\u2019s other physiological effects are numerous. A 2012 meta-analysis examined 14 randomized trials, containing 1068 patients, and assessed berberine\u2019s efficacy in numerous biometrics, including resting plasma glucose levels, plasma insulin levels, body weight, body mass index, and reduction in fat mass and serum triglycerides and total cholesterol content. The aggregate data suggests that berberine performs at least as well as oral hypoglycemic agents (such as metformin) in these key metrics.<\/p>
Boron is a mineral present in multiple fruits, vegetables, tubers, and drinking water, though it is not widely accepted as either an essential mineral or essential vitamin. Boron affects a few physiological functions in the body, particularly modulating hormonal homeostasis.<\/p>
While still emerging, recent data suggest that boron may play a role in potentiating free testosterone levels in even healthy men. In one study, otherwise healthy men were provided an acute serving of boron and had their free and total levels of testosterone, alone with SHBG (sex hormone binding globulin) and DHT (dihydrotestosterone) levels measured. In that study, total testosterone was unaltered, but free testosterone was enhanced by 14%, while SHBG was significantly (9%) reduced.<\/p>
Choline is an essential nutrient involved in numerous metabolic pathways, including DNA regulation and repair, protein function, and metabolism. Perhaps most importantly, the critical neurotransmitter acetylcholine is produced directly from free choline via cholinergic neurons. Acetylcholine is then responsible for a number of functions itself, most crucially as the compound which induces muscular contraction, and as the neuromodulator partially responsible for modulating risk\/reward, arousal, and enhancing memory.<\/p>

As mentioned at the beginning, the Tiger analogy breaks down a bit, since we are not pure carnivores. We bring a significant number of carbs into the game \u2013 or at least you should be if you\u2019re trying to maximize growth. Carbs tie in to insulin which, I\u2019m sure you\u2019ve heard, is an extremely anabolic hormone. But insulin is also something to be feared, as shuttling nutrients into our muscle also comes with the caveat of your hungry, hungry hippo...err, fat cells trying to greedily suck up nutrients as well. Here we use an inducible receptor \u2013 which means it isn\u2019t bound to the membrane necessarily but must be signaled to emerge from deep inside the cell out to the membrane to interact with, mostly, glucose. This receptor is called a GLUT4 receptor (side note, we will touch briefly on the GLUT1 receptor, also a glucose transporter, but we know a bit less about it)<\/p>
GLUT4 \u2013 Berberine cause dramatic increases in expression of the GLUT4 transporter, and the effects seems to be dominant in muscle cells rather than fat cells.<\/p>
By increasing GLUT4 activity in muscle cells, especially when timed after some glycogen depletion and a hearty carb intake post workout, we can stuff dem dere muscle cells right back up to full, creating an environment ripe for muscle growth. Remember, when the body is given ample nutrients, it interprets this as you are evolutionarily successful, and will be rewarded with \u201caccess\u201d to more resources (muscle, strength, stamina...and downstream from that \u2013 better choice of mates (giggity).<\/p>
Berberine sounds amazing! Why not just load up? Well, grasshopper, berberine has some drawbacks. For one, it has absolutely dismal oral absorption. And if you try to make up for that by taking more, well make sure you stay close to a bathroom. Intestinal irritation comes with high doses. Instead what I have done is dialed in a Berberine-Nicotinamide (more in a second) Cocrystal. Recall that cocrystals can be formed by dissolving 2 or more compounds in a commonly effective solvent, applying some initial heat, and then allowing the solvent to evaporate. As the fluid volume decreases, our compounds are forced together where physics does its thing \u2013 and they form a crystalline lattice, which changes the physical properties such as solubility and absorption. In this case, the crystals are beautiful and very effective.<\/p>
Berberine has also been shown to inhibit the Devil Incarnate of bodybuilding\u2026.MYOSTATIN (boo, boo, hiss). It also inhibits expression of SMAD 2\/3. See the BMP writeup, but SMAD 2\/3 are the bad ones for muscle building. Using Slintensity in conjunction with BMP will amplify the effects. Yes way.<\/p>
Berberine also has the unfortunate property of increasing expression of Atrogin-1, a highly catabolic\/atrophic pathway. So while you are waiting like Christmas Eve for Santa to bring you muscle, Krampus shows up to undo it. So what did I do about that? Good questions<\/p>
Puerarin comes from the a few plants, but luckily for your wallet it can be easily extracted from Kudzu. Corosolic Acid? From Banaba? YAWN...I thought you were innovative. Well, hang on. In the previous version of Slintensity, we first address the role of the TGR5 Bile Acid receptor using Obacunone. Obacunone was co-extracted with Berberine from the Phellodendron Cortex plant. Unfortunately, as I moved on with my experiments using purified Berberine cocrystals, my engineers were unable to separate the two, which means Obacunone was no longer an option. Having moved through several other compound, each with steep absorption problems and an even steeper price tag, I wound up finding an obscure study comparing TGR5 activators. The most potent in the study was none other than Corosolic Acid. But, at much higher dosage than was being used. This took a ton of work to get made, and still isn\u2019t cheap, but fortunately CA has excellent absorption.<\/p>
TGR5 \u2013 lets revisit our Tiger Analogy. Remember, without receptors and other sensors the body is blind and stupid. Nutrient sensors in particular signal to the body WHAT and HOW MUCH you are eating. There are carb and protein sensors, but what macronutrient is left behind? Yep\u2026.fat. One of the ways the body is able to digest fat is using bile acids. What does that have to do with our Tiger you might ask? Well, when a successful kill is made our tiger begins his feast \u2013 and it ain\u2019t frosted flakes he\u2019s eating. Flesh, blood, muscle \u2013 protein, and quite a bit of fat. So, corosolic acid. We needed a target range of 40-50mg for ideal activation of TGR5. Since the most common CA Banaba extract is 1%, there was no way to make it viable. That is why I used a 10%. I\u2019m working on an even more potent custom extraction for the future.<\/p>

Agmatine Sulfate is one of the few ingredients that has been shown to have the capability to increase GLUT-4 transportation and mRNA expression in skeletal muscle, making it a prime ingredient to help ensure that glucose transport to muscle cells is favored over fat cells (10). It achieves this through an indirect action on beta-endorphin secretion which, in much the same mechanism as exercise, promotes glut-4 translocation into skeletal muscle through activation of imidazoline I2A receptors. (10, 18)<\/p>
Berberine HCI is one of the most effective ingredients currently available for the purpose of reducing blood sugar. While Berberine's exact method of action is still currently under investigation, it is purported that its ability to activate AMPK is a driver for the regulation of carbohydrate through AMPK's ability to promote glycogen synthesis and increase glucose uptake in insulin sensitive cells, thus removing glucose from the blood faster for immediate oxidization or stored as glycogen for later use as fuel. Also, due to AMPK's ability to promote insulin sensitivity and glucose uptake in a method independent of insulin itself, Berberine may have the capability to promote glucose uptake in cells that have become insulin resistant, thereby effectively clearing the blood of glucose in even the most difficult of environments. Bitter Melon has several active constituents that promote glucose from clearing the blood, with these being charantin, vicine, and polypeptide-p.(13,14) Each one of these exerts their hypoglycemic effect via different physiological modes of action, ranging from stimulation of skeletal muscle utilization to preservation islet B cells (cells integral to proper insulin functioning) thus governing most of the process that underpins blood glucose clearing and helps preserve the integrity of the process. Of importance is the role of protecting Islet B cell health, whereby destruction of these cells results in a lack of insulin production. Bitter Melon protects against this down-regulating the cytokines responsible for islet cell death, namely interleukin-6, tumor necrosis factor a, and nuclear factor kappa b. Banaba leaf, most notably its primary active ingredient, Corsolic Acid, has been reported to decrease blood sugar levels within 60 minutes in human subjects through its ability to increase cellular uptake of glucose. However it isn't just its ability to reduce blood glucose that is of value, it is also its ability to assist with weight loss. In a study that tested a combination of Banaba, Bitter Melon, Gymnema, and Bioperine on weight loss, subjects lost an average of 6 kilograms over a 12 week period. (13)<\/p>
In various other human trials, Banaba extracted for Corsolic Acid has been noted to decrease blood glucose by up to 30% in a matter of hours, even when compared to placebo. Its proposed mode of action lies within its ability to stimulate Glut 4 translocation and induce insulin receptor phosphorylation, both of which promote the clearance of glucose from the blood. (14)<\/p>
Formulations that include Gymnemic Acids have been used as a useful tool in the fight against obesity. The proposed method of action is through its ability to delay glucose absorption in the blood, thus blocking the over secretion of insulin which may under normal circumstances dump glucose into adipose tissue. (5, 16) Gymnemic Acids have a very similar structure to that of glucose which enables them to compete for the same receptor locations in the intestine, effectively delaying the absorption of glucose into the blood. Gymnema is also particularly useful at curbing sugar cravings due to its ability to fill taste receptors on the tongue, thereby preventing the action of sugar molecules attempting to stimulate the same receptors. (16)<\/p>
Other mechanisms by which Gymnema is proposed to work include increasing the secretion of insulin, promoting regeneration of islet cells, and increasing the utilization of glucose. (5, 16)<\/p>
Alpha Lipoic Acid (ALA) is a mitochondrial compound heavily involved in energy metabolism. It has various benefits across a myriad of biological processes, including but not limited to neuroprotection, cholinergic transmission, dopaminergic transmission, cholesterol transport, and energy expenditure. (8, 15) It also has a range of benefits pertaining to glucose uptake and AMPK activation, outlining a similar method of action to Berberine. In the murine model, Alpha Lipoic Acid is able to significantly mediate gene expression key rate limiting enzymes involved in glucose metabolism, which is able to increase the rate of glycogen production in the liver. (8, 11, 15, 17)<\/p>
Na-R-ALA is the sodium version of the R isomer for Alpha Lipoic Acid which is the most stable and bioavailable form of ALA currently available. It works through the same mechanisms as Alpha Lipoic Acid, but on a much larger scale. Na-R-ALA is most commonly known for its ability to produce dramatic muscle pumps, resulting from increased glucose uptake in skeletal muscle.<\/p>
Saffron is one of the most underrated glucose disposal agents currently available. AMPK on the other hand plays a vital role in glucose and lipid metabolism. Both pathways increase phosphorylation of mitogen-activated kinase (MAPK) which participates in full activation of insulin-stimulated glucose uptake via GLUT-4. Saffron actives both pathways, hence its effectiveness in clearing blood glucose into skeletal muscle and, where too much glucose is shuttled, it is able to facilitate its preference to glycogen storage over adipose tissue storage.
